Reproduction, epigenesis and development

 

One of the conducting thread that permits evolution from the oocyte up to embryonic stem (ES) cells is epigenetic remodeling, particularly via the parental imprinting process.
Imprints establishment occurs during gametogenesis and is maintained in somatic cells through embryonic development and post-natal life.
Our team examines the role of different perturbators on genomic imprinting ressetting during gametogenesis and on genomic imprinting expression in stem cells.
Part of the team is involved in developping new processes to preserve women fertility. This includes ovary cryopreservation and grafting and ovocytes vitrification that are validated according to the integrity of genomic imprints.
The other part of the team will evaluate the stability of the imprint in stem cell lines in collaboration with C Dehay and P Savatier Teams

- Evaluation of the  human immature oocyte vitrification, followed by IVM, on the methylation profile of H19 and KCNQ1OT1 imprinting centres.

 

- Methylation of the pluripotency factor promoters  in human embryos at 2-4 cell   stage, progressive or blocked in their development.

Subject: Vitrification of whole sheep ovary

I work on whole sheep ovary cryopreservation through vitrification. The purpose is to conserve the integrity and functionality of the ovary at very low temperature. The aim of this study is to validate a technic to preserve woman fertility prior to cancer tratement. Cryopreserved ovaries will be grafted to the patients following cure.
